Procedures/protocols
developed for germination of immature embryos of epiphytic
and terrestrial orchids species/hybrids
-
Sporadic
germination of seeds in most of the species.
-
The
days for swelling of embryo varied from 4 days to 115 days
and for globule constitution 6 to 120 days.
-
The
time taken among the embryos of various developmental stages
of Cymbidium crosses for swelling varied from 35-92 days and
PLB’s development 75-128 days.
-
Protocorm
morphology and subsequent development of PLB’s was quite
similar in all the species.
-
Supplementation
of BAP resulted in proliferation of protocorms and addition
of activated charcoal enhanced shoot growth besides root
development.
-
Among
various media tried, Murashige and Skoog’s and Nitsch
found to be ideal for many of the species for germination,
proliferation and differentiation.
-
Supplementation
of paclobutrazol to the pre hardening media helped in
development of quality plantlets with dark green and thicker
leaves and better survival.
-
Triacontanol
also enhanced the plantlet growth besides better root
development.
-
Among
the various potting media, equal proportion of coco pith and
sand or coco pith alone found suitable for growth of micro
propagated plantlets of Cymbidium hybrids.
